چکیده

The binding of aminoacyl transfer ribonucleic acid @RNA) to ribosomes in the presence of corresponding messenger RNA (specific binding) is compared with that in the absence of messenger RNA (nonspecific binding). Specific binding is much more sensitive to tetracycline than nonspecific binding, and the former is similar to, but not identical with, the latter in its response to changes in Mg++ concentration. On the other hand, trypsin treatment of ribosomes abolishes both nonspecific and specific binding of aminoacyl-tRNA to the ribosomes in a parallel fashion, indicating that ribosomal proteins are involved in the binding of aminoacyltRNA in general. Aminoacyl-tRNA bound in the absence of messenger RNA can be partially, but not totally, chased from ribosomes by the addition of messenger RNA which does not contain the code word for the ammo acid. For this removal of aminoacyl-tRNA by unrelated messenger RNA, unbound, specific aminoacyl-tRNA coded for by the messenger must be present. Thus, the removal is an exchange reaction with the incoming specific aminoacyl-tRNA, and the addition of messenger alone in the absence of incoming specific aminoacyl-tRNA does not remove the nonspecifically bound aminoacyl-tRNA.

Studies on the Formation of Transfer Ribonucleic Acid-Ribosome Complexes VII. THE ROLE OF THE 3’-HYDROXYL-TERMINAL END OF TRANSFER RIBONUCLEIC ACID FOR INTERACTION WITH RIBOSOMES AND RIBOSOMAL SUBUNITS*

The ability of altered transfer ribonucleic acid to compete with binding of [14C]aminoacyland [l*C]tRNA was used as an estimate of its capacity to bind to ribosomes. Transfer ribonucleic acid with the terminal 3’-hydroxyl rA, ,C,A, and ,C,C,A enzymatically removed could bind to 70 S ribosomes with progressively lower efficiency; in contrast, binding to 30 S subunits was unimpaired. Transfer rib...

Reaction of ribosome-bound peptidyl transfer ribonucleic acid with aminoacyl transfer ribonucleic acid or puromycin.

Polylysyl transfer ribonucleic acid, free of lysyl-tRNA, has been isolated and its characteristics in an Escherichiu cofi polypeptide-synthesizing system in vitro have been studied. Ribosomes obtained from frozen E. coli cells were washed in 0.5 M NH&l and assayed at MgCla concentrations of at least 0.01 M. Binding of this peptidyl-tRNA to ribosomes occurs in the presence of templates containin...

A method which uses cyanogen bromide for the quantitative conversion of 4-thiouridine to uridine has been developed. The general nature and the specificity of this reaction has been studied with thiouridine residues present in a hexanucleotide isolated from enzymic digests of Escherichia coli tyrosine transfer ribonucleic acid (tRNA), and also with those present in intact E. coli tyrosine and f...
